Abstract

A total of five traditional rice plants (Oryza sativa L.) and five modem cultivated rice plants were studied using Random Amplified Polymorphic DNA (RAPD) markers to evaluate their genetic diversity. The RAPD markers employed were OPAA-04, OPB-07, OPC-06, OPC-20, OPF-17, OPH-13, OPK-12, RAPD-R2, RAPD-R3 and RAPD-R5. Based on the analysis, there were 57 bands produced and 55 bands among them were polymorphic. The polymorphism among the paddy varieties was high, 96.49%. An UPGMA dendrogram computed based on the cluster analysis of genetic distance showed the paddy samples had a significant amount of genetic variation Among all the RAPD markers, OPK-12 is one of the markers which can be used to detect the blast resistance gene in paddy. As a result, four samples of paddy, Sachupak, Mayang Ebos 136, Pongsu Seribu IT and Malinja were detected and considered had the potential to carry the blast resistance gene which link to the OPK-12 marker.
