Abstract

The aim of the study was to determine the growth behaviour of Lactobacillus addophilus (LA-S) and 8ifidobacterium laros (88-12) in banana and pineapple purees. Level of fruitto-water ratio and supplementation (yeast extract and L-cysteine) for both fruit purees were determined and their effect towards the kinetic growth of L. acidophilus and 8. lactis was observed. Total titratable acidity, pH and total soluble solids were measured during the fermentation. Results revealed that both non-supplemented fruit purees favoured the growth of L. acidophilus and 8. lactis, 90% of pineapple puree and 70% of banana puree were selected as media to determine the kinetic growth of probiotics. In non-supplemented pineapple puree, the maximum growth of B. lactis was slightly higher than L. acldophilus but B. lactis required relative longer time to achieve the maximum growth. Similarly, L. acidophilustook 6 hours shorter than 8. lacticto get to the peak of growth in non-supplemented banana puree. The pH of non-supplemented media inoculated with L. acidophilus always showed a higher reduction than same media inoculated with B. lactic The effect of yeast extract towards the growth of L. addophilus and 8. lactis was significantly higher than L-cysteine supplementation. Growth of L. acidophilus and B. lactic in fruit purees supplemented with 4-6% of yeast extract was significantly higher than their growth in milk. Symbiotic growth effect seems to exist between L. acidophilus and B. lactis in pineapple medium, but not in banana medium. Incorporation of B. lactic in pineapple medium appeared to increase the survivability of both problotics significantly (p<0.05). However, the viable count of B. lactis in mixed cultures was lower than the single culture during 8 - 48 hours of fermentation in banana medium. As conclusion, banana and pineapple medium are capable to support the growth of L. acldophllus and 8. lactis (with or without supplementation) and potentially to become an alternative low cost growth media for culturing L. acldophilus and B. lactis.
