Abstract

A genetic relationship study was made correlating pineapples from three locations in Sabah. The study aimed at developing DNA fingerprints that were able to be used to characterize high marketable pineapples in order of location in Sabah. DNA was extracted from pineapples originating from Tamparuli, Sipitang and Babagon; a total of 40 samples per location. PCR was performed prior DNA extraction using DALP primer pairs. DNA fingerprints of pineapples were produced using three DALP forward primers, DALP232, DALP233 and DALP234. Each forward primer was paired with the universal M13 reverse primer, DALPR. PCR products were separated using Polyacrylamide gel electrophoresis for greater resolution, followed by the calculation of genetic similarity and genetic distance using RAPDistance software. In the final analysis a dendogram was constructed by the obtained genetic similarities and distances index. The primer DALP232IDALPR produced 47 bands, of which 8 were polymorphic. Primers DALP233IDALPR and DALP234IDALPR each produced 37 and 29 bands respectively, where the former exhibited 7 polymorphic bands while the latter 6 polymorphic bands. The primer DALP232IDALPR had a polymorphism percentage of 17.02%, whereas DALP233IDALPR 18.92% while DALP234IDALPR was 20.69%. The dendogram extrapolated from the genetic similarity and distances index data obtained showed that the Gula Batu pineapple variety from Sipitang and Tarnparuli are more closely related genetically between each other compared to the pineapples from Babagon.
