Abstract

Analysis on total bacterial count (TBC) and total Vibrio count (TVC) was conducted for the period of seven months (June to December 2004) at the Borneo Marine Research Institute Finfish Hatchery. Seawater samples from the Borneo Marine Research Institute (BMRI) jetty, seawater after filtration, plankton culture cum brackish tilapia culture tank brood fish tank and larval rearing tank were collected following standard method analyzed for the TBC and TVC. The water samples were spread onto two solid culture media; Tryptone Soya Agar (TSA) and Thiosulfat Citrate Bile Salt Sucrose Agar (TCBS). Plates were incubated for 48 hours at 25 °C before the colony-forming unit (CFU) were observed. Although the values of CFU for TBC and TVC fluctuate according to month, it remains high as compare to the value recorded for seawater in an open system. T -test analysis was conducted to compare means of TBC and TVC in the samples taken from different facilities system in the hatchery as well as in open water where the seawater was pumped from. The analysis showed that both total bacterial count (p>0.05) and total Vibrio count (p<0.05) in the hatchery facilities (broodstock tanks, larval rearing tanks and plankton culture tanks) were significantly higher than the available number of viable bacteria in sea water. Physical water parameters (dissolved oxygen, pH, temperature and salinity) did not significantly affect the bacterial growth. The study suggested that analysis of other biochemical water quality parameters such as ammonia, nitrites and nitrates are urgently needed as these nutrients have been described as growth promoters among marine bacteria and stressors to marine organisms, especially fish.
