Abstract

An axenic culture is a situation in which only one species is present and it is uncontaminated by organisms of other species. To obtain the axenic culture of Pyrodinium hahamense var. compressum. Prorocentrum micans and Gymnodinium calenatum, the antibiotic treatment (mixture of three antibiotics: penicillin G, streptomycin and chloramphenicol with the ratio of 10:5:1), ultrasonic treatment, potassium tellurite (K₂TeO₃) treatment, washing with ethanol and spray plating method were used in this study. After each treatment, the microalgae were checked for bacteria-free by using the marine agar media 2216. For P. bahamense and P. micans, 0.5m1 of the antibiotic mixture, 10p,glml of potassium tellurite and 30 minutes of sonication followed by centrifuge for 5 minutes at 6000 rpm able to reduced the amount of bacteria, but some of the algae cannot survive. 1 m1 of antibiotic mixture able to eliminate all the bacteria but the algae cannot survive. By washing with 70% ethanol, almost 100010 of the cultures were free from bacteria contamination. When washing with 60% ethanol, 52% of the cultures were free from bacteria contamination. For G. calenalum, 0.5ml of antibiotic mixture able to reduce the amount of bacteria, but the algae cannot survive. G. calenatum cannot treat with ultrasonic treatment and washing with ethanol because the cell burst after these treatments. This may be due to its structure, which is the unamored dinoflagellate. P. bahomense, P. micans and G. calenatum cannot grow on L10 and f/20 agar media. In conclusion, all the methods have been used were unable to obtain axenic culture.
