Abstract

Transfonnation of Labisia pumila var. pumila (Kacip Fatimah) was perfonned by using two different strains of Agrobacterium rhizogenes which were TR105 and A4GUS. Type of explants, type of strains and the best OD reading of Agrobacterium growth were the three objectives studied in this experiment. All the explants had been wounded before inoculated in Agrobacterium culture with OD reading 0.4, 0.5 and 0.6 for TR105 strain and 0.5, 0.7 and 0.9 for A4GUS strain for 30 minutes. All the explants were removed onto co-cultivation medium for 6 days followed by transferring of explants onto fresh MS with double vitamin medium for regeneration phase. In this experiment, the explants that had been ninoculated in sterilized distilled water and cultured on MS with double vitamio were used as control. Three trials of transformation were carried out for each type of explants with the same 00 reading of each strain. For Histochemical test, Agrobacterium A4GUS was used. The result of this experiment was, there was overgrowth problems of Agrobaclerium strain A4GUS occurred 00 all explants that have been inoculated with OD reading OD₆₀₀ 0.5 and 0D₆₀₀ 0.7 after co-cultivation period although all the explants had been washed with antibiotic for forth time. Test of Histochemical could not be done on all the explants with overgrowth problems but for the explants that were inoculated at OD₆₀₀ 0.9, this test was carried out and showed negative result There was no hairy root formation found on explants that were transformed with TRI05 strain.
