Abstract

Black Bone Silky Chicken (BBSC) is one of the indigenous chicken breed. BBSC is high in nutritious value but the growth rate is slow as compare to commercial broiler. Commercial broiler was a hybrid derive from White Plymoth Rock and Conish Game for meat production with fast growth rate. The objective was to study the gene diversity of BBSC and broiler using GHR-Intron 2 primer. A number of 30 broiler DNA samples and 42 BBSC DNA samples were obtained and screened by RFLP-PCR. The genotypes frequency of GHR-Intron 2 for A�A�' A�A₂ and A₂A₂ were 0.0417, 0.0833 and 0.7500, respectively. Alleles frequency for allele A� was 0.0952 and allele A₂ was 0.9084. The expected heterozygosity was 0.1528 and observed heterozygosity was 0.0833. The observe heterozygosity was smaller than the expected heterozygosity indicates that inbreeding depression in BBSC population. Whereas broiler population was not performed random mating due breeding selection program. The genetic similarity was 0.9745 between the population and the genetic distance was low (0.0258). A dendogram was constructed using genetiC distance and the neighbour-joining method. The result showed that the genetiC constitution between broiler and BBSC was less divergence.
