Abstract

There has been an increasing number of food borne outbreaks occurring in beef burger. However, there has been limited number of publications investigate on pathogenic bacteria in beef burgers in Malaysia. The objective of this study was to identify pathogenic bacteria in beef burgers locally manufactured in Malaysia. Bacteria were isolated from three samples of beef burger from different Malaysian manufacturer. The samples were cultured on MacConkey Agar and Mannitol Salt Agar to differentiate between Gram-negative and Gram-positive bacteria. Morphological studies were done to identify the colony characteristics of the bacteria. Eight out of 24 strains of bacteria were selected for biochemical tests (API 20E and API for staph). The strains were identified as S. auricularis, S. hominis, S. schleiferi, S. haemolyticus, Shigella sonne~ Enterobader sakazakil, Salmonella spp., and Pseudomonas oryzihabitans. The samples were screened for antibiotic resistance using antimicrobial susceptibility test. Every strain showed different resistance pattern since antimicrobial resistance quickly developed as a natural effect of the ability of bacteria to adapt antimicrobial agents. Molecular Identification was carried out to further reconfirm the Identity of the bacteria strain by amplification of 16s rRNA gene. DNA sequencing showed that one of the strains was belong to Enterobader spp. As a conclusion, there is a possibility of Identifying pathogenic bacteria in beef burger isolated from local manufacturers.
