Abstract
This study was conducted to determine the occurrence of plasmids in the 22 strains of Antartic bacteria which came from 12 genus; flavobacterium, Pseudosomonas, Microbacteliaceae, Aermoicrobium, Rhodococcus. Dyadobacter, Brevundimonas. Cryobacterium, Methylibium, Sphingomonas, Bacillus, and Arthrobacter. There were 15 bacteria samples isolated from Crater Lake, Deception Island while the other 7 isolates came from Estrella Lake, King George Island. The medium used were LS, R2A, and Marine. Three DNA extraction methods were used by using phenol: chloroform extraction method, CTAB extraction method, and alkaline lysis method. Based on the gel electrophoresis result, CTAB extraction method had successfully extracted the DNA from all bacteria samples. Gram staining was conducted to ensure the purity of the bacteria samples where there were 13 Gram-positive bacteria and 9 Gram-negative bacteria. However, this study showed that no plasmids detected from the 22 Antarctic strains.
