Abstract
Pedobacter cryoconitis BGS are psychrophiles that are isolated from cold environment and capable of proliferating and growing well at low temperature. Their cellular products have found a broad spectrum of application including food, medical as well as bioremediation. Cellular product derived from an organism greatly influenced by high cell density cultivation that is associated with the suitable growth medium supplemented with the right nutrient components. However, nutritional requirement can contribute to the production cost and the bioprocess economics. Up to date, there has been no published report on the optimization of growth medium of pedobacter cryoconitis BGS for cost effective production. In the present study, a preliminary screening on commercially available media was conducted to identify potential nutrient components. Tryptic Soy broth produced the highest cell density and four medium components were selected (tryptone, glucose, NaCl and K2HP04) along with three additional nutrients (yeast extracts, MgSO4 and NH4CI) for Plackett-Burman-based statistical screening to determine the significant medium components affecting cell density of pedobacter cryoconitis. Results show that tryptone, yeast extract and NH4CI give positive effects and influence most on the production of cell density. Response surface methodology with Central composite design was adopted to optimize the three nutrient components. Statistical analysis of the data shows an 83% model fitting results predicted optimum cell density of 7 .5 O.D when using 23.7 g/L of tryptone, 8.8 g/L of yeast extract and 0.7 g/L of NH4CI. However, the observed value (6.8 O.D) was lower than expected. Nevertheless, the optimized growth medium are more cost effective compared to TSB.
