Labisia pumila is a traditional herb used by Malay women in postnatal health care. This study involves the phytochemical screening and biology activities of the plant roots crude extract of petroleum ether, chloroform and 1-butanol. The crude extract yield by each solvent is 1.61%, 0.29% and 1.28% respectively. The phytochemical screening showed the presence of saponins steroidal and cardiac glycosides in all extracts. While the chloroform extract exhibit the presence of alkaloid, flavanoid and antraquinone bioactive groups. The chloroform extract had 3.44 relative toxicity compared with potassium dichromate. The relative antioxidant activity of the chloroform and 1-butanol extracts compared to BHT are 1.246 and 1.283 respectively. Allelopathic test showed that 1-butanol extract had a strong seed germination inhibitor property. There is no positive result obtains from MAPK kinase and MAPK kinase phosphatase inhibitor test. A column chromatography separation of chloroform extract using a mixed solvent of benzene, chloroform and ethyl acetate at 3: 1: 1 ratio had isolated a pure orange compound (K5) and being re-crystallized with n-hexane. The compound IR spectrum analyses showed a hydroxyl and C=C group also sp2 and spa hybrid -CH.
