Abstract

Synthetic antioxidants such as butylated hydroxytoluene (BHT) and butylated hydroxylanisole (BHA) are currently used as food additives, and many plant species have similar antioxidant potentials as these synthetics. In this research, bioactive components of Morinda citrifolia L. fruit was extracted by conventional solvent extraction and supercritical fluid extraction. Microcapsules containing Morinda citrifolia L. - microparticles having antioxidants were produced by spray-drying using different proportions of K-carrageenan and maltodexrin as binding materials. The results were evaluated based on the process parameters of spray drying on microencapsulation yield, particle size, DPPH scavenging activity, total phenolic content and total flavonoid, for different volume ratio of Mcore/Mwall and different inlet temperatures of microencapsulated powder. From the results a ratio of 1:2 at 90° C was found to be the optimal operating spray drying condition for Kcarrageenan and maltodextrin for conventional solvent extraction extract. The results of SEM and particle size analyzer showed that spray dried microcapsules had a regular spherical shape with a diameter of 2-5 IJm for K-carrageenan and maltodextrin. However, spray dried supercritical fluid extract (SFE) of M. citrifolia L. showed higher antioxidant activity than conventional solvent extraction extract. The highest antioxidant activity, total phenolic content and total flavonoid were found at a 1:2 ratio of Mcore/Mwall at 90°C for maltodextrin (SFE with cosolvent). Under these optimal operating conditions, the antioxidant activity, total phenolic content and total flavonoid content were found to be 32.3%, 56 mg of TAE/g of SDP and 60 mg of CE/g of SDP, respectively. The microencapsulation yield, particle size and moisture content of the spray-dried fruit extract produced using maltodextrin as the adjuvant were found to be 38.5%, 2.27 µm and 0.133 respectively. Process optimization was also done for conventional solvent extraction extract of M. citrifolia L. using RSM. This study will be helpful in the pharmaceutical application
