Abstract

Groupers are important for aquaculture and the fisheries industry world wide. The complete mtDNA sequence of the E.lanceolatus from Sabah had been determined. The mtDNA sequences consists of 16649 bp comprising 13 genes and 22 tRNAs and 2 rRNAs. The gene organization and gene content in E. lanceolatus conformed to that observed in other veterbrates. Most of the genes were encoded on the H-strand except for NADH6 and eight other tRNAs as found in other veterbrates. The reading frames of four pairs of coding genes were found to be overlapping: ATP8 & ATP6; ATP6 & COIII; NADH4L & NADH4; NADHS & NADH6. The codon usage and the base composition had been detailed. The putative structure of the origin of the L-strand replication is proposed. The sequence of the control region and the conserved sequences block were identified. In the NJ and UPGMA analyse, the complete genome and 12 coding genes of E. lanceolatus from Sabah positioned in the clade including E lanceolatus from Taiwan, E coioldes and E bruneus. A Similarity search between E lanceolatus from Sabah and Taiwan showed high homologies in nucleotide sequences except for the NADHS gene. Most of genes mutated non-synonymously indicating that positive selection is occuring and providing a fitness advantage in population. The mtDNA of E lanceolatus did exhibit variation in certain genes. Six of the primers were able to cross amplify in E fuscoguttatus and a clear patterns of maternal inheritance was evident in the F1 hybrid. Haplotype analysis has a potential application in broodstock management programs.
