Abstract

The actinomycete strain H7393 is potential inhibitor for yis mutant Mkk1p386. The focus of this research is to continue Cheah.H. Y.(2003) research's on strain H7393 which done in her thesis ' Isolation of Actinomycetes from mangrove in Sabah and screening for inhibitor against eukaryotic signal transduction'. Strains H7393 were grown in mannitolpeptone medium aerobically to produce secondary metabolites. The secondary metabolites then extract by acetone. The acetones extract of actinomycete H7393 showing inhibition in MAPK Kinase screening. H7393 extract is then freeze dry into powder and is dissolved in distilled water to test on thin layer chromatography. Purpose of thin layer chromatography is to separate the compounds in the strain actinomycete H7393 . We found that the solvent system comprising methanol and water at ratio 4: 1 can separated the compound clearly with its Rf value for the first spot is between 0.6 and 0.8. while the Rf value for the second spot is between 0.3 and 0.5. In the screening system for human GSK-3β inhibitor, a total of 114 extract strain actinomycete had been tested on the screening system for human GSK-3β inhibitor and yeast Bull inhibitor. The result for the both screening system, the extract of strain H7667 has been found as inhibitor of human GSK-3β . While the extract of strain H11110 and strain H8985 was found as both the inhibitor of human GSK-3β and yeast Bull. The strain H8934, H7944, H8604, H8996, Hl1013 and H11ll] has been found as toxic to both the screening system of human GSK-3β and yeast Bull
