Abstract
Mannnase is an enzyme known to degrade hemicellulolytic component and has been used in gas and well simulation processing of kraft pulp. food processing and additive in detergent In Malaysia. mannanase is used to treat Palm Kernel Cake (PKC) based poultry and ruminant feed for the removal of mannan and subsequently increasing feed conversion of ruminant and poultry. The aim of this study is to isolate some bacteria which produce mannanase that are active at normal or low temperatures to treat PKC. Bacteria from soil sample around Sabah and Antarctica were screened for mannanase activities using Remazol Brilliant Blue-Guar Gum and Locust Bean Gum as substrates. Twenty five mesophilic and 3 psychrophilic isolates with mannanase activities were isolated. Gram staining was conducted on all the bacteria isolates to observe their morphologies. All isolates exhibited bacillus-like morphology. To diffentiate the bacteria, Randomly Amplified Polymorphic DNA (RAPD) was carried out and the DNA fingerprinting analysis revealed that 17 out of the 25 mesophilic bacteria isolated were different Five mesophilic bacterial strains (Y1I. Y2. Y16. Y18 and A7) and three Antarctica strains were identified based on their 16S rRNA gene sequences. The 16S rRNA gene was partially sequenced and the results revealed that isolates Y1 and Y2 have high homology with uncultured bacteria, isolates, Y18, Y16 and A7 were from the genus Paenibacillus while all the three Antarctic strains, showed high homology to bacteria from the genus Bacillus.
