Abstract
Isolation of high quality RNA from mature banana fruit tissue is difficult due to high levels of polysaccharides and other substances that interfere when using conventional procedures for RNA isolation. These substances do not only decrease the yield but the quality of RNA is almost unusable. Isolation of RNA is very easy to contaminate by ribonuclease. Two methods had been used in my research were CTAB/NaCI method and phcnol/chloroform/isoamylalcohol. These methods were suitable for isolation RNA from ripening banana fruit. To compare which is more suitable, the measurement by spectrophotometer and analysis of gel electrophoresis had been carried out. The CTAB/NaCI method is more effective procedure because it yielded 8.25 μg of total RNA per g tissue, while phenol/chloroform isoamylalcohol method yielded 3.18 pg of total RNA per g tissue. The integrity of the RNA which obtained from CTAB/NaCI method is also better than phenoUchloroform/isoamylalcohol method. The A2SA280 ratio for RNA sample from CTAB/NaCI method is 2.0 while 1.7 for phenoUchloroform/isoamylalcohol method.
