Abstract
The rare of Dimorphorchis lowii with the special for its appearance and scent of dimorphic flowers is endemic to Borneo. An efficient in vitro methodology for mass propagation of the orchid is needed to produce plants for commercial production and to increase the populations in its natural habitat. In this study, leaf explants were cultured on Mitra basal medium supplemented with various concentration of cytokinin (BAP) and auxin (NAA), individually or/and in combination under continuous darkness. The results showed that after 8 weeks of culture, the cut end of basal leaf explants had the highest competence to form embryoid-like structures (ELSs) than the cut end of leaf tips. The combination of 6 mg l�¹ NAA with either 1 mg l�¹ or 4 mg l�¹ BAP, and 6 mg l�¹ BAP alone showed the highest percentage of explants producing ELSs (26%, 20 %, and 20 % respectively). Regardless of the concentrations (1, 2,4, and 6 mg l�¹), addition of NAA alone into the media showed the highest percentage of necrotic explants (47 % - 73 %). For the growth regulators-free medium, 85 % of the explants became necrotic after 8 weeks of culture.
