Abstract
Gelatin is a yellowish granule, odourless and flavourless protein that is used in food processing industry and also pharmaceutical industry. However, the acceptability of gelatine products by consumers greatly depends on the animal sources of the gelatine. As such, porcine and bovine gelatine are the two primary sources used in the large industries but limited acceptance because of religious prohibitions and potential zoonotic threats. Although, identification of gelatine source used is important for gelatine products but it is very challenging due to the low content of DNA in the sample and strong interaction between extracted DNA and the nonlysed gelatine residue. Therefore, there is a need for a rapid method to detect the porcine DNA from the pharmaceutical capsule for the purpose of gelatine source identifications. In this research project, various pharmaceutical capsules were used as a source material to detect the presence of porcine (pork) DNA by using different types of DNA extraction methods. This way then confirm and verify by Polymerase Chain Reaction (PCR) analysis. DNA was successfully extracted from the capsule with high purity DNA using Sodium Dodecyl Sulfat (SDS) compare three other methods, CTAB method, DNeasy Blood and Tissue Kit, and EasyPure® Food DNA kit. PPA6 primers which targets the mtATP6 gene was then used to amplify porcine DNA using Conventional Polymerase Chain Reaction (PCR) assay. It successfully amplified the gene and this was observed on 83 bp DNA band upon agarose gel electrophoresis. This method of species identification particularly for porcine DNA detection is very useful to verify whether the gelatins from pharmaceutical capsules or gelatin-containing food products are adulterated with porcine ingredients.
