Abstract
Cancers as a group account for approximately 13% of all deaths each year leading to high morbidity and mortality. In recent years, a number of natural products have been found to inhibit cancer growth. In search of a better natural based cancer treatment, a study on the effectiveness of a plant called Ruellia tuberosa was performed. Stems and leaves of Rue/lia tuberosa were extracted and underwent solvent-solvent fractionation to separate active compounds according to polarity. These fractions were used in three different tests: MTT assay to evaluate the ability to inhibit cell growth, DPPH assay to evaluate the antioxidant contents and DNA fragmentation test to detect the presence of apoptosis process. The antioxidant activities of the different fractions tested decreased in the order of ME>EaF>WtF>HxF>CfF in stem extract and EaF>WtF>ME>CfF>HxF for leaf extract according to the DPPH free radical-scavenging assay. Among ten extract fractions (methanol, water, chloroform, hexane and ethyl acetate extract of stem and leaf) of this plant, ethyl acetate extract of the leaf showed highest inhibitory effect on MCF-7 cancer cell growth when tested using MTT assay. However, when the DNA of the cancer cell that treated with the same extract is isolated and run in gel electrophoresis, there were no fragmentation shown, indicating the absence of apoptosis process. This study suggests that ethyl acetate extract of Ruellia tuberosa contain high antioxidant and can inhibit cancer cell growth in in vitro. Further study need to be done on its ability to induce apoptosis process in cancer cells. The results provide useful information on the pharmacological activities associated with cancer cells of this traditional folk remedy.
