Abstract

The hexane, methanolic (dichloromethane, ether. ethyl acetate, butanol and water) and ether: ethyl acetate extracts from bambangan seeds were tested on their total phenolics content by using Folin-Ciocalteu. antioxidant activity by using 2,2-Diphenyl-1- picrylhydrazyl (DPPH) free radical scavenging, ferric reducing antioxidant power (FRAP) and beta-carotene bleaching methods and antimicrobial activity were tested by disk diffusion assay, broth dilution method and time-kill study. The extracts shown a total phenolics content ranging from 0.30-366.11 mg/ml garlic acid equivalent (GAE) where methanolic butanol extracts gave the highest phenolics content while hexane extract was the lowest. Methanolic ether. ethyl acetate extracts shown the most potent antiradical activity (EC50 = 0.02 mg/ml). All extracts found to be stronger radical scavenger compared to the standard, butylated hydroxytoluene (BHT) except hexane extracts. Except for hexane and rnethanolic dichloromethane extracts, others extracts all have the higher FRAP antioxidant activity compared to BHT standard. Among the extracts, methanolic ether: ethyl acetate extracts gave the highest antioxidant capacity (2702.7 µM/I FeS04.7H20 Equivalent at concentration of 0.25mg/ml) and its antioxidant activity can not be determined at the concentration after 0.25mg/ml. For beta-carotene bleaching methods, the IC50 of the methanolic ether: ethyl acetate, butanol and water extracts were 0.96,0.08 and 0.12 mglml respectively. The standard (BHT) can not reach IC50 at test concentration. Other extracts of hexane, methanolic dichloromethane and crude ether: ethyl acetate can not be detected due to the insolubility of the extracts in water. For antibacterial activity, methanolic ether: ethyl acetate and methanolic butanol partitions gave the most potent inhibiting properties on the extend. The inhibition diameter zones for both extracts are significantly (p < 0.05) stronger than other extracts toward Staphylococcus aureus and Salmonella typhimurium while methanolic ether: ethyl acetate has the greatest inhibition on Salmonella typhimurium and Staphylococcus aureus at 25.84±0.63mm and 21 .07±0.71 mm, respectively. Methanolic butanol gave a total of 22.16±0.5 3mm on Staphylococcus aureus and 2S.84±0.63mm on Salmonella typhimurium. Other extracts had shown a comparable results to the commercialized antibiotics. For minimum inhibitory concentration, methanolic butanol gave the strongest inhibition towards the test microorganism at the range of 15.625-7.8125mg/ml for Salmonella typhimurium, Sfapgylococcus aureus and Bacillus cereus and 31.25ft 15.625mg/ml for Pseudomonas aeruginosa. Minimum inhibition concentration (MIC) and minimum bactericidal concentration (MBC) of butanol extracts against Staphylococcus aureus were 9.3mg/ml and 10.mglml respectively meanwhile Salmonella typhimurium were 10.8mg/ml and 12.3mg/ml respectively. Decimal reduction time (D-value) for methanolic butanol extracts against Staphylococcus aureus was 6.49 hours while Salmonella fyphimrium was 6.92 hours. The data presented were comparatively higher than commercialized standard for antioxidant and antimicrobes. So, the antimicrobial and antioxidant activity of bambangan seeds were potential as a new substitution but required further study.
