Abstract
Vibrio spp. are among the most important fish pathogens that cause bacterial disease in cultured fish such as grouper. Several of these species are also harmful to humans, which include Vibrio cholerae, Vibrio vulnificus, Vibrio alginolylicus, and Vibrio parahaemolyticus. Six isolates of bacteria were taken from diseased marbled grouper (Epinephelus .fuscoguttatus): TgVibJR1, TgVibJR2, TgVibJR3, TgVibJR4, TgVibJR5 and TgVibJR6. Gram staining, motility tests and other conventional biochemical tests were perfonned as well as the usage of API-20E test kit to identify these bacteria. TIrree species were identified, Vibrio alginolyticus as TgVibJR1 and TgVibJR2 isolates, Vibrio parahaemolyticus as for TgVibJR4, and Shewanella putrefaciens as the remaining strains. The effects of ten commercial antibiotics were tested through an antibiotic susceptibility test. Results indicate that all strains have total susceptibi]jty towards 50 µg furazolidone, 300 µg nitrofurantoin and 25 µg streptomycin whereas low concentrations of the vibriostatic agent 0129 and ampicillin at 10 µg are ineffective against. all strains. Vibrio alginolyticus was selected as the test bacterium in the viru1ence test, using marbled grouper fingerlings measuring 3 - 4 inches in length. The pathogenic nature of Vibrio alginolyticus was confirmed when after 60 hours of the initial inoculation with the bacteria, the fishes at all concentrations were found dead as compared to the control. Bacterial swabs taken from the kidney of the dead fish formed growths on TSA and TCBS agar. Colonies were observed as round and yellow in colour.
