Abstract

The purpose of this study was to determine the Minimum Inhibitory Concentration of crude extracts from Phyllanthus niruri (Dukung anak) and Cymbopogon citratus (Lemongrass) against food borne pathogens such as Staphylococcus aureus, Escherichia coli 0157: H7, Listeria monocytogenes and Salmonella typhimurium. The herb samples were extracted by using water, methanol and ethanol; steam distillation was used to extract essential oil. Essential oil was dissolved in 0.01% of Tween 80. Antibacterial activity of the herb extracts were tested using disk diffusion method and then tested in liquid medium to determine minimum inhibitory concentration (MIC). The results of disk diffusion method showed S. aureus was strong inhibited by flavonoid extract, methanol extract, water extract and essential oil. All of the extracts from C. citratus leave were strong active against E. coli. The extracts that were most active against S. typhimurium were methanol extract, essential oil and flavonoid extract. L. monocytogenes was resistant to all extractions except essential oil with 32.75 mm zone of inhibition. Methanol and flavonoid extract from P. niruri, water extract and essential oil of C. citratus leave had MICs of 31.25 mg/ml, 12.5 mg/ml, 250 mg/ml and 2 mg/ml against S. aureus. The lowest MIC against E. coli, L. monocytogenes was 0.5 mg/ml essential oil. Methanol extract, essential oil of C. citratus leave and flavonoid of C. citratus stem had MICs of 25 mg/ml, 0.5 mg/ml and 15.625 mg/ml against S. typhimurium. In 24 hours, S. aureus had totally killed by methanol extract (62.5mg/ml) and flavonoid extract (25 mg/ml); E. coli killed by methanol extract (25 mg/ml) from P. niruri; methanol extract (25 mg/ml), flavonoid extract (62.5 mg/ml), ethanol extract (25 mg/ml) from C. citratus leave. L. monocytogenes had killed by 2 mg/ml essential oil in 6 hours. S. typhimurium had totally killed in 6 hours by essential oil at concentration 2 mg/ml. Among these extractions, essential oil is most effective to inhibit the growth of pathogen. Therefore, these extractions could serve as potential antibacterial agents to inhibit pathogen growth in food.
