Abstract
Dengue virus is a type of harmful virus transmitted primarify' by Aedes mosquitos which causes a global health problem due to the lack of ideal vaccine and also ineffective vector control program. Therefore, a fast, sensitive and reriable method is needed for the detection of dengue virus. In this study, an electrochemical DNA sensor was developed based on the modified gold electrode (AuE) with nanocomposite membrane of chitosan which consisted of multi waD carbon nanotubes and gold nanoparticles for the specific detection of dengue-3 serotype (DENV-3). These components have good conductivity and biocompatib[rt:y, in which the combination of these nanomaterials can effectivefy' increase the electron-transfer and current responses. ImmobiUzation process was constructed based on thiol-gold interaction while the hybrk:tization process was performed by targeting DENV-3 specific sequence. The electrochemical DNA sensor was found to be optimized under these conditions; 6. 5 pH, 10 s interaction time, 0.25 V/s scan rate and 25° c temperature. The morphological characteristics of multiwaD carbon nanotube and gold nanoparticles were uniformfy' distributed in the CHIT nanocomposite membrane. Under optimal conditions, Differential Pulse Voltammetry was used with different concentrations of target DNA on the range from 1 x 104until 1 x 108 • The modified electrode has shown a good sensitivity towards the target dengue serotype-3 DNA and was able to detect a very low concentration of 1x108 target DENV-3 compared to the unmodified electrode. The developed DNA sensor is expected to provide an earfy' detection of dengue virus during ( 1-3) days since appearance of symptom.
